Modulation of the Antidiabetic Effect of Glimepiride by Diazepam in Diabetic Rats

چکیده
Anxiolytic drugs may influence glycemic control in diabetic subjects, so the present study was conducted to investigate the effect of glimepiride, a commonly used oral hypoglycemic, diazepam, a commonly used anxiolytic, and their combination in STZ-induced diabetic rats. Diabetes was induced by streptozotpcin (50 mg/kg i.p.). Rats were divided into 5 groups namely: normal control, diabetic control, glimepiride (10 mg/kg p.o.), diazepam (5 mg/kg i.p.) or combination of both glimepiride and diazepam, respectively. All groups received daily treatments for 2 weeks including the normal group which received 1% Tween 80. Diazepam significantly improved the effect of glimepiride on the levels of serum glucose, insulin, C-peptide and liver glycogen content. In addition, combination of diazepam with glimepiride significantly improved the effect of the latter on oxidative stress biomarkers including serum lipid peroxides, blood glutathione levels and blood superoxide dismutase activity of diabetic rats. In conclusion, the present study revealed that diazepam increased the antidiabetic and the antioxidant actions of glimepiride which may be of considerable value in the treatment of diabetes mellitus where both reduction of associated anxiety and tight glycemic control are needed.

Effect of Sitagliptin and Glimepiride on Glucose Homeostasis and cAMP Levels in Peripheral Tissues of HFD/STZ Diabetic Rats
Introduction: T2DM is a group of metabolic disorders manifested by hyperglycemia as a result of insulin insufficiency and/or resistance. The main goal of antidiabetic therapies is to lower glucose levels, and therefore prevent development of diabetes complications. DPP-4 inhibitors (e.g. sitagliptin) are relatively new antidiabetic drugs which inhibit the activity of DPP-4 enzyme and therefore ...

Pancreatic β-cells dysfunction and impairment of insulin action usually leads to hyperglycemia. Punica granatum L. is a well-known traditional herbal remedy in Iran due to its positive effects on ameliorating blood glucose homeostasis. In this study, we aimed to investigate and compare the pomegranate fruits aqueous extract (PE) on glucose and lipid metabolisms in normal and Alloxan-induced dia...

Type 2 diabetes mellitus (T2DM) is a group of metabolic disorders characterized by hyperglycemia owing to insulin resistance and/or insulin deficiency. Current theories of T2DM pathophysiology include a decline in β-cells function, a defect in insulin signaling pathways, and a dysregulation of secretory function of adipocytes. This study aimed to investigate the effect of different antidiabetic...

UNLABELLED As a nutritional supplement, coenzyme Q10 (CoQ10) was tested previously in several models of diabetes and/or insulin resistance (IR); however, its exact mechanisms have not been profoundly explicated. Hence, the objective of this work is to verify some of the possible mechanisms that underlie its therapeutic efficacy. Moreover, the study aimed to assess the potential modulatory effec...
